Study on the process conditions of enzymatic hydrolysis of porcine hemoglobin

In the abstract experiment, neutral protease was used to hydrolyze porcine hemoglobin. The effects of hydrolysis temperature, pH value, enzyme addition amount, substrate concentration and hydrolysis time on the hydrolysis effect were studied by single factor test. Through orthogonal test, the optimal hydrolysis conditions were as follows: temperature 55 °C, pH 7.5, enzyme addition 5 000 U/g protein, substrate concentration 10%, hydrolysis time 3 h, degree of hydrolysis under these conditions The soluble nitrogen content (SN-TCA index) of 22.03% and trichloroacetic acid was 63.83%.

However, due to the common red hemoglobin powder, the color is red, the blood smell is heavier, the smell is bad, the palatability is poor, the sensory quality is affected, and the digestive enzymes lacking red blood cells and the amino acid composition imbalance in the intestinal tract of the animal (leucine and lysine) Higher acid content and lower levels of isoleucine and methionine limit the widespread use in animals. Therefore, researching suitable hemoglobin powder processing technology, expanding the amount of addition and scope of use, can not only make up for the shortage of protein resources in China, but also can obtain huge economic benefits and reduce environmental pollution.
The protease hydrolysis technology can degrade hemoglobin into polypeptides, small peptides and amino acids which are beneficial to the digestion and absorption of the animal body. In particular, hemoglobin rich in hemoglobin can be used as an ideal iron supplement blood source to produce high quality functional feed materials. In this paper, we use neutral protease to enzymatically hydrolyze pig hemoglobin, study the enzymatic hydrolysis parameters of the best hemoglobin powder, and provide some practical reference for further deep processing of hemoglobin powder.
